It's pretty obvious to me that if they don't join forces they'll just end up on Sylar's list. I think the villans will only work together for a small goal, not the overall goal. And Wolverina is the only one who could with stand a megaton atomic detonation...right? I like the theory that there are actually 3 parties involved.
Right now it's the Heroes, Claire's Dad and Voodoo Man, and maybe someone else?
Here's my take on "save the cheerleader, save the world".
1) Either they have to band together, and it's just a quest or...
2) Future Hiro knows that Sylar would be invincible if he eats Claires brain, or...
3) (my favourite:) It's all really simple. If Claire dies i.e. is not saved, Daddy / Sylar flips out (because he's actually trying to take care of her) and in his rage of the loss of his daugher, nukes NYC.
Personally I think Peter is to beat Sylar. Claire is adpopted, her father must know her abilities already. I think he is powerless, and I do think someone's got something, maybe Sylar gains powers through the naught brain eats. Not sure. I deffinatly see Niki turning bad though, I dont think she's good. Its deffinatly more then a split personallity, she seems to gain strenth in UBER-Bad@$$ form.

i think there is more at stake than just NY being blown up. otherwise it would say SAVE THE CHEERLEADER, SAVE NEW YORK! i think the bomb will have a dangerous biological agent, like a virus or something. cheerleader is weak-ass, has no power other than regeneration/healing. maybe they need something from within her to heal the sick people of the world when the virus spreads. the indian geneticist dude can use his skills finally. it's like the Terminator storyline, where it wasn't about stopping the catastrophe, but to keep the right people alive and together so that they can fight the bad guys in the future. hiro is the man to bring them together!
Any thoughts that perhaps Sylar is Claire's father? I do not think her adoptive dad is Sylar. Since the cop shot who we think is Sylar and he rose afterwards, that hints at existing regenerative powers already.

Oh and Jazzy, you consider her "weak-ass"? Wow, I though of her as in pretty good shape physically, and seeing as she can not stay dead, I consider her a power house! Imagine the drive and determination that could give someone, knowing they can not get permanantly hurt.
by weak i mean her value in battle situations. unless she trains her fighting skills or gets an adamantium transfusion like wolverine, she is dependent on the others to protect her. her special ability is undeniably awesome and obviously she is important, but still the others must SAVE her. maybe later the writers will maker her triumph as an individual. claire is the classic 'small/weak person who can't really fight, but has a huge impact on outcome of story' type character. LOTR anyone?
LOTR indeed. Could HRG be Sylar? Interesting, though I doubt it.
I'm interested in seeing what happens when Peter is around multiple heroes at once... Maybe THAT is the key to beating Sylar.

And the brain-eating M.O. from Sylar...I wonder if he starts targeting heroes one by one. Theory: The destruction of NY/Nuke/Whatever could be a ruse to get these heroes to band together, possible to take them all out at once? The only two that saw it are Isaac and Hiro, and it's possible that it could be avoided just by them NOT doing anything about it. By saving the cheerleader, they could end up NOT going to NY and therefore not being there to be nuked, stopping the nuke from happening because they aren't there to target.
